In vitro Effects of Exogenous Fibrolytic Enzymes on Rumen Fermentation of Wild Guinea Grass (Panicum maximum)

چکیده مقاله:
This study aimed to evaluate the potential of commercial enzymes cellulase (CE), xylanase (XY) and the combination of CE + XY to improve rumen fermentation of Guinea grass ecotype ‘A’ (Panicum maximum). The experiment was carried out in a randomized complete block design. In vitro incubations were performed with four doses of individual enzymes and their combinations (1:1 from each enzyme) as 50, 100, 150 and 200 µL enzymes with 500 mg substrate. In vitro gas production (IVGP) was measured at 4 h intervals. At the end of 24 h incubation in vitro rumen dry matter disappearance (IVRDMD), ammonia nitrogen (NH3-N), protozoa population and volatile fatty acid (VFA) were estimated. Supplementation with CE, XY and CE + XY significantly enhanced IVGP (control: 38.54 mL; CET1: 50.06 mL; XYT1: 54.27 mL and CET1 + XYT1: 52.77 mL) and IVRDMD (control: 46.78%; CET1: 51.21%; XYT1: 51.53% and CE1 + XY1: 52.64%). The rumen NH3-N production was significantly increased (P

Use of exogenous fibrolytic enzymes to enhance in vitro fermentation of alfalfa hay and corn silage.
Two in vitro experiments were performed to identify promising exogenous fibrolytic enzyme products (EFE) and optimum dose rates (DR) for improving the degradation of alfalfa hay and corn silage. The relationship between enzymatic activity and fermentation responses was examined to identify optimum formulations. In experiment 1, 5 EFE containing mainly endoglucanase and xylanase activities, with...
متن کاملInfluence of supplementary fibrolytic enzymes on the fermentation of corn and grass silages by mixed ruminal microorganisms in vitro.
This study was done to determine the effectiveness of supplementary enzymes at increasing the fiber digestion by ruminal microorganisms and to assess whether enzyme activity limits the rate of fiber digestion in ruminal digesta. In vitro comparisons of enzyme activities in two feed enzyme preparations (A and B) with enzyme activities extracted from ruminal fluid indicated that the addition of f...
متن کاملInfluence of Fibrolytic Enzymes on the in vitro Hydrolysis and Fermentation of Different Types of Roughages Treatment
The effects of pre-treating different types of roughages with alkali on the efficacy of exogenous fibrolytic enzymes for improving their digestibility were studied in vitroin factorial arrangement 4 × 3 × 5 (enzyme, treatment and roughage types). Two fibrolytic feed enzymes novozyme (N) and celloclast (C) their combination (N+C) were evaluated for their potential to improve in vitrodegradation ...
متن کاملThe Effect of Native Grass Substitution Using Jengkol (Archidendron jiringa) Peel and Leaves Powder on in vitro Rumen Fermentation
The effect of substituting native grass with jengkol (Archidendron jiringa) by-product on fermentation characteristics, rumen microbial profile, methane production, and hydrogen balance using in vitro method was investigated. Seven treatments (different composition of native grass, jengkol peel, jengkol leaves, and concentrate) with five replications in a block randomized desi...

In this study, 20 different isolates of lactic acid bacteria (LAB) were physicochemically screened for the ability to grow at low pH and produce lactic acid. The best isolates were selected for fermentation of Panicum maximum in order to produce silage. The grass was harvested at 6 weeks re-growth and ensiled at 15, 30, 45 and 60 days. Samples were taken on each of these days for microbial chan...
